Abstract
:
During inflammation, neutrophils are rapidly recruited at sites of acute infection and dominate the initial influx of leukocytes. Phosphoinositide 3-kinase (PI3K) is intracellular signalling pathways have been found to be of particular importance in the recruitment, activation and survival of neutrophils. This study aimed to investigate the impact of Interleukin-8 (IL-8) inhibitor on intracellular signaling enzymes as respiratory burst responses . The study included 10 normal, healthy volunteers (age range: 18-50) not suffering from any systematic disease or taken any medication. Blood was collected on heparinzed tube then the neutrophils were isolated and counted. Superoxide anion generation was measured as the superoxide dismutase (SOD) inhibited reduction of ferricytochrome c. The enzyme linked immunosorbent assay system (ELISA) was used to read the different concentrations of IL-8 and Phosphoinositide 3-kinase gamma (PI3Kγ) II inhibitors. Results obtained showed that, the optimal concentration of IL-8 -induced superoxide production from human peripheral blood neutrophils was 10-7μM and the optimal concentration of PI3Kγ II inhibitors which can inhibit O2- generation was 10-7μM. By using t-test the comparison between the controls and the different concentrations of IL-8 with or without SOD showed that there are no significant differences between neither the negative nor the positive. The comparisons between the negative and positive controls and the different concentrations of PI3Kγ inhibitor II without SOD using showed that there are significant differences between the positive control and the10-5 (P= 0.023), 10-6 (P= 0.027), 10-7 (P= 0.039), and 10-8 (P= 0.041) concentrations, with SOD there was significant differences between the positive control and 10-7 (P= 0.033) concentration of the. In conclusion, IL-8-induced the respiratory burst in human peripheral blood neutrophils that inhibited partially by PI3Kγ inhibitor II.
