Abstract : Biodegradation of poly 3-hydroxybutyrate (bioplastics) by Streptomyces bacterium By Ghofran Nezar Abd-Almouti Redwan Abstract Actinomycetes are filamentous Gram-positive bacteria mostly found in soil and have important roles in breaking dawn of the organic materials and polymers. About 30 actinomycete isolates were obtained from saline soil collected from Jeddah on starch nitrate agar using serial dilution method. All isolates were screened for poly-3-hydroxybutyrate (PHB) degradation on agar and broth media containing PHB as carbon source. The isolate GhM7 was the most active isolate in PHB degradation which was identified as species belong to genus Streptomyces using morphological and molecular identification in addition to examination using light and scanning microscopes. It was identified as Streptomyces sp. GhM7. The best degradation was recorded at 40ᵒC after 4 days of incubation in minimal medium containing no glucose or yeast extract as additional carbon and nitrogen sources, respectively. Acidic or alkaline pHs like pH 6, 7.5 and 8.0 enhancing PHB degradation. PHB concentrations affect degradation process, 0.5 g of PHB showed the best enzyme production. The bacterium was grown in the broth medium at the best growth conditions and the filtrate was collected and depolymerase was precipitated, purified and characterized. The maximium enzyme activity was at 40ᵒC and pH 7. Additionally, the presence of heavy metals affect significantly the enzyme activity. In conclusion, the species belonging to the genus Streptomyces showed excellent activities in PHB degradation and improving growth conditions enhanced the degradation process and depolymerase enzyme production.
